Description

 The MoClo Baculo Toolkit is a modular cloning system designed to simplify and speed up the generation of multigene expression constructs for the baculovirus expression system. The toolkit is based on Golden Gate Assembly, using Type IIS restriction enzymes (BsaI or BsmBI). Plasmids are typically assembled from multiple DNA parts in a one-pot, one-step reaction without PCR or Gibson assembly. The system supports flexible tagging of proteins dedicated for expression and purification, and provides compatibility with the biGBac Kit and MoClo-YTK.

The toolkit includes all the plasmids required for domesticating parts and using them to build baculovirus expression vectors of up to six genes each. The number of proteins that are expressed from a single vector can be increased up to 30 through compatibility with the biGBac Kit. 

Therefore, users who intend to use the MoClo Baculo Toolkit to build baculovirus expression vectors of seven or more proteins are advised to obtain the biGBac Kit in addition to the MoClo Baculo Toolkit. Users who wish to use their MoClo Baculo-compatible open reading frames and tags to build yeast expression vectors should obtain the MoClo-YTK in addition to the MoClo Baculo Toolkit.
